Applications : For specifically measuring histone H3-K23 acetylation using a variety of mammalian cells (human, mouse, etc.) including fresh and frozen tissues, cultured adherent and suspension cells.
Storage : Upon receipt, store F3, F4 and standard control at –20°C. Store all other components at 4°C away from light. The components of the kit should be stable for 6 months when stored properly.Note: Check if buffers F1 and F2 contain salt precipitates before using. If so, warm (at room temperature or 37°C) and shake the buffers until the salts are redissolved.

For Research Use Only. Not intended for any clinical use. No products from Creative BioMart may be resold, modified for resale or used to manufacture commercial products without prior written approval from Creative BioMart.

Ask a question
Histone acetylation mechanism? 12/02/2020

Histone acetylation and deacetylation are essential parts of gene regulation. These reactions are typically catalysed by enzymes with "histone acetyltransferase" (HAT) activity. Acetylation is the process where an acetyl functional group is transferred from one molecule (in this case, acetyl coenzyme A) to another. Deacetylation is simply the reverse reaction where an acetyl group is removed from a molecule.Acetylation removes the positive charge on the histones, thereby decreasing the interaction of the N termini of histones with the negatively charged phosphate groups of DNA. As a consequence, the condensed chromatin is transformed into a more relaxed structure that is associated with greater levels of gene transcription.

Can samples flash frozen in liquid nitrogen and stored at -80˚C be tested? 11/04/2019

Yes. But the activity from frozen tissues may be slightly lower than that obtained from fresh tissues.

What method/protocol can I use to quantify proteins/histones after the histone extraction procedure? 10/12/2018

We recommend using any standard protein quantification assay such as a Bradford assay for histone quantification.

Can tissues or cell pellets be frozen prior to histone extraction? 09/03/2017

Yes, it is possible for tissue or cell pellets to be frozen before extracting histone proteins.

What equipment will be used in the experiment? 12/13/2016

You will need a fluorescent microplate reader and plate.

Can I use tissue samples lysed in RIPA buffer in my experiments? 08/24/2014

RIPA buffer would not be compatible with the 10X Lysis Buffer. Please use the lysis buffer in the kit.
