Product Overview:
Rabbit Monoclonal Antibody to Human Tumor Necrosis Factor Receptor Superfamily, Member 21
Antigen Description:
Human death receptor-6 ( DR6 ) is identified as a member of TNF receptor superfamily. This type I transmembrane receptor possesses four extracellular cysteine-rich motifs and a cytoplasmic death domain. DR6 is expressed in most human tissues and the highest levels were detected in heart, brain, placenta, pancreas, thymus, lymph node and several non-lymphoid cancer cell lines. DR6 interacts with the adaptor protein TRADD and mediates signal transduction through its death domain, and expression of DR6 in mammalian cells induces activation of both NF-kappaB and JNK and cell apoptosis. DR6 knockout mice have enhanced CD4+ T cell proliferation and Th2 cytokine production, suggested that DR6 serves as an important regulatory molecule in T-helper cell activation, and is involved in inflammation and immune regulation. In addition, DR6 is uniquely cleaved from the cell surface of tumor cell lines by the membrane-associated matrix metalloproteinase (MMP)-14, which is associated with tumor malignancy.
Specificity:
Human DR6 / TNFRSF21. No cross-reactivity with human cell lysate ( 293 cell line ) in WB and ELISA.
Immunogen:
Recombinant human DR6 protein
Host:
Rabbit
Isotype:
Rabbit IgG
Species:
Human
Clone:
A002
Applications:
Western blot; ELISA
Dilution:
Western blot: This antibody can be used at 1 - 2 μg/mL with the appropriate secondary reagents to detect human DR6 in WB. Using a DAB detection system, the detection limit for human DR6 is approximately 10 ng/lane under non-reducing conditions and 5 ng/lane under reducing conditionsDirect ELISA: This antibody can be used at 0.1 - 0.2 μg/mL with the appropriate secondary reagents to detect human DR6. The detection limit for human DR6 is approximately 0.039 ng/well
Preparation:
This antibody was obtained from a rabbit immunized with purified, human cell-derived, recombinant human TNFRSF21
Format:
0.2 μm filtered solution in PBS with 5% trehalose
Storage:
This antibody can be stored at 2-8°C for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20 to -70 °C. Preservative-Free.Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.
