|Description:||Tetanus toxin is a potent neurotoxin that is taken up by presynaptic terminals at the neuromuscular junction, travels retrogradely within motor neurons and is transported transsynaptically to spinal cord interneurons. Upon entering the motor neuron, tetanus toxin associates with lipid rafts, is incorporated into non-acidified vesicles, sorted in a Rab 5/7 dependent pathway and transported to the cell body. The tetanus toxin is trancytosed so that it enters upstream inhibitory neurons where it cleaves VAMP-2, inhibiting neurotransmitter release. Tetanus toxin inhibits neurotransmitter release with great potency and selectivity, cleaving a bond in vesicle-associated membrane protein-2 (VAMP-2) or synaptobrevin-2, thereby preventing the fusion of synaptic vesicles with the presynaptic membrane. Tetanus toxoid is currently used as a vaccine to produce immunity to tetanus, the disease caused by Clostridium tetani. Additionally, tetanus toxoid functions as a carrier of conjugated polysaccharides in the development of vaccines against encapsulated bacterial pathogens.|
|Form:||Each vial, when reconstituted with 0.2 ml of water, contains 0.1 mg of tetanolysin in 0.02 M sodium phosphate, 0.1 M NaCl, pH 7.2. Handle the product gently, do not vortex.|
|Bio-activity:||Hemolytic activity of the tetanolysin was determined with fresh rabbit red blood cells (RBC's). One hemolytic unit (HU) is the quantity of tetanolysin in 1 ml which causes 50% hemolysis of a RBC suspension at 5x1 09 cells/ml at 37 centigrade within 40 minutes. For this lot, one HU is equivalent to 6 ng of tetanolysis, giving a specific activity of approximately 170 HU/ug. Since each cell type exhibits a different sensitivity, testing activity against a given cell type by using toxin at 0.1, 1.0, 10 and 100 HU/ml is highly recommended.|
|Endotoxin:||The endotoxin content, determined using a kinetic chromagenic LAL assay, is 300 EU/mg.|
Tetanolysin is a very potent toxin: microgram quantities are lethal for mice, rabbits and other laboratory animals. The lethal (cardiotoxic) and cytolytic properties are suppressed by oxidants. The biological properties are irreversibly lost in the presence of low concentrations of cholesterol and other 3[3 hydroxysterols which interfere with toxin binding to the target cells.
|Storage:||Tetanolysin is supplied as'a lyophilized powder, sealed under vacuum. Store at 2-8 centigrade prior to reconstitution.|
