Ask a questionThe combined knockdown of CALB2A and CALB2B genes in zebrafish results in severe hydrocephalus, defective axial curvature, and pericardial edema. These phenotypic consequences suggest that CALB2A and CALB2B play crucial roles in processes related to brain development, body curvature, and heart function in zebrafish.
The gene expression of these calcium-binding proteins in the central nervous system (CNS) and peripheral nervous system (PNS) during the early embryonic development of zebrafish has been demonstrated using the "whole mount in situ hybridization" study
These proteins have been well-studied in the CNS of different vertebrate groups, including fish. They are often found in the central nervous system, intestine, kidney, and pancreas of higher classes of vertebrates, such as amphibians and birds.
A morpholino-based knockdown strategy was employed to generate zebrafish mutants with reduced CALB2A and CALB2B gene expression. When CALB2A and CALB2B were individually knocked down, no significant phenotypes were observed. However, simultaneous knockdown of both genes resulted in the development of several notable developmental defects.
The observed phenotype in the Calb2a and Calb2b combined mutants is similar to the acerebellar (ace) mutants, which also lack the cerebellum and have a disorganized MHB boundary. This correlation suggests that the Calb2a and Calb2b genes may have a role in cerebellum development similar to that of the ace mutants.
