    MKK3 and MKK6 are two closely related dual-specificity protein kinases that activate p38 MAP kinase. MKK3 and MKK6 both phosphorylate and activate p38 MAP kinase at its activation site, Thr-Gly-Tyr, but do not phosphorylate or activate Erk1/2 or SAPK/JNK.

What is MAP2K6 protein?

MAP2K6 (Mitogen-Activated Protein Kinase Kinase 6) is a protein coding gene which situated on the long arm of chromosome 17 at locus 17q24. This gene encodes a member of the dual specificity protein kinase family, which functions as a mitogen-activated protein (MAP) kinase kinase. The MAP2K6 protein is consisted of 334 amino acids and its molecular mass is approximately 37.5 kDa. The secreted recombinant human MKK6 consists of 336 amino acids and predicts a molecular mass of 37.6 KDa. The apparent molecular mass of the protein is approximately 37 KDa in SDS-PAGE under reducing conditions due to glycosylation.

What is the function of MAP2K6 protein?

MAP kinases, also known as extracellular signal-regulated kinases (ERKs), act as an integration point for multiple biochemical signals. MAP2K6 (Mitogen activated protein kinase 6) is a mitogen-activated protein kinase kinase, which belongs to the MAPK signaling pathway. Its main function is to regulate biological processes such as cell growth, differentiation and apoptosis. Specifically, MAP2K6 can phosphorylate its downstream MAPK kinases, such as MAPK3 and MAPK6, thereby activating the activity of these kinases. These kinases further phosphorylate other proteins, including transcription factors, cytoskeletal proteins, and other signaling molecules, thereby regulating the physiological function of cells. In addition, MAP2K6 can also influence cell survival and death by regulating cell cycle and apoptosis.

MAP2K6 related Signaling pathways

MAP2K6 is involved in the MAPK signaling pathway, which includes a three-stage signaling process: MAPK, MAPK kinase (MEK or MKK), and MAPK kinase kinase (MEKK or MKKK). These three kinases can be activated sequentially, and together regulate a variety of important physiological and pathological effects of cells.
MAP2K6 can also open heterochromatin by phosphorylation of Gatad2b, improve the histone acetylation modification in the promoter region of pluripotent genes, and finally improve the expression of pluripotent genes and promote the acquisition of pluripotent genes.

MAP2K6 Related Diseases

Mutations in MAP2K6 gene are associated with the occurrence and development of a variety of tumors, such as breast cancer, colorectal cancer, gastric cancer and so on. In addition, abnormal expression or mutation of MAP2K6 gene can lead to neurodegenerative diseases, such as Alzheimer's disease and Parkinson's disease. It is also associated with some other diseases, such as cardiovascular diseases, diabetes, cirrhosis and so on.

Bioapplications of MAP2K6

In some tumor cells, MAP2K6 expression is associated with tumor aggressiveness and poor prognosis. Therefore, MAP2K6 is considered a potential therapeutic target that could be used to develop new anticancer drugs. In addition, MAP2K6 opens heterochromatin by phosphorylation of Gatad2b, improves histone acetylation modification in the promoter region of the pluripotent gene, and ultimately improves the expression of the pluripotent gene and promotes the acquisition of pluripotency, which is also an ideal model for studying epigenetic regulation.
