What is AZGP1 protein?

AZGP1 ( alpha-2-glycoprotein 1, zinc-binding) gene is a protein coding gene which situated on the long arm of chromosome 7 at locus 7q22. This gene encodes a protein called Zinc-alpha-2-glycoprotein (ZAG), also known as AZGP1. This protein is found in body fluids such as serum, sweat, and seminal and breast cyst fluids. It is identical in amino acid sequence to tumor-derived lipid mobilizing factor (LMF), a protein associated with the dramatic loss of adipose body stores in cancer cachexia, and has been shown to stimulate lipolysis by adipocytes in vivo and in vitro. ZAG exhibits a class I major histocompatibility complex (MHC) fold but is a soluble protein rather than being anchored to plasma membranes and does not associate with alpha-2-microglobulin in humans. This protein is consisted of 298 amino acids and is approximately 34.2 kDa.

What is the Function of AZGP1 protein?

AZGP1 is a multidisciplinary protein, which is secreted in various body fluids. The AZGP1 plays roles in lipolysis, regulation of metabolism, cell proliferation and differentiation, regulation of melanin synthesis, cell adhesion, and immunoregulation.

It can bind to DNA and interact with transcription factors, thereby influencing the transcription of these genes. One of the well-known functions of AZGP1 is its involvement in lipid metabolism and adipose tissue regulation. It has been shown to promote lipolysis (the breakdown of fats) in adipocytes and enhance the release of fatty acids into the bloodstream for energy production.

AZGP1 Related Signaling Pathway

The P-AKT/GLUT4 signaling pathway is an important downstream pathway for insulin action, and AZGP1 can alter the sensitivity of adipocytes to glucose and further affect blood glucose control by affecting this signaling pathway. It was also found that Renvastinib can activate AZGP1 by increasing the acetylation level of H3K27Ac in AZGP1 promoter region, and regulate TGF-β1/Smad3 signaling pathway in an AZGP1-dependent manner, thereby inhibiting liver cancer cell migration and invasion (ICC-EMT).

AZGP1 Related Diseases

AZGP1 is expressed at reduced levels in some tumors and is therefore considered a tumor suppressor protein. It has been found that the expression level of AZGP1 is closely related to tumor growth, metastasis and prognosis in a variety of cancers, such as prostate cancer and lung cancer. AZGP1 may also be involved in the occurrence and development of metabolic diseases such as insulin resistance and obesity.

Bioapplications of AZGP1

AZGP1 has a promising application in metabolic regulation, cancer diagnosis and treatment, and immune regulation. For example, AZGP1 may be a potential key target for Lenvatinib in the treatment of hepatocellular carcinoma (ICC).
