Description: Defensins (alpha and beta) are cationic peptides with a broad spectrum of antimicrobial activity that comprise an important arm of the innate immune system. The alpha-defensins which include NP-1, NP-2 and NP-3, are distinguished from the beta-defensins by the pairing of their three disulfide bonds. In addition to antimicrobial activity, NP-1 exhibits chemotactic activity on dendritic cells. NP-1 is expressed as the C-terminal portion of an inactive precursor protein, which also contains a 19 amino acid N-terminal signal sequence and a 45 amino acid polypeptide. NP-1 contains a six-cysteine motif that forms three intra-molecular disulfide bonds.

Purity: >95% by SDS-Page and HPLC analysis. Endotoxin level is less than 0.1ng per ug (1EU/ug) of human0 NP-1.
Reconstitution: The lyophilized human NP-1 is soluble in sterile DI water . The material can be reconstituted to a maximum concentration of 1mg/ml in water. Allow to set at least 30 minutes at 4 °C. Tighten cap, mix gently by inversion. If desired, can centrifuge for 1-2 minutes at 1000RPM to concentrate material in vial. This solution can be diluted into water or other buffered solutions or stored at 4 °C for up to one week or at -20 °C for future use.
Note: If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >50ug/ml.
Reactivity: This recombinant human NP-1 can be used at between 1-10ng/ml to chemoattract immature dendritic ce.
Storage: The lyophilized powder, though stable at room temperature for a few weeks but is best stored desiccated at -20°C. Reconstituted NP-1 should be stored in working aliquots at -20 °C. Avoid Frequent Freeze-thaw Cycles.
