Description : HDAC8 phosphorylated in vivo by PKA results in decreased activity, which leads to the hyperacetylation of histones H3 and H4. HDAC8 may play a role in one of the most frequent types of acute myeloid leukemia (AML). The active deacetylase is useful in the study of HDAC8 regulation and for inhibitor screening.

Recommended Reaction Conditions : 137 mM NaCl, 25 mM Tris-HCl, 2.7 mM KCl, 1 mM MgCl2, pH 8.0, 100 µM fluorescent substrate [Arg-His-Lys(Ac)-Lys(Ac)], 100 µg/ml BSA, 15 ng/ml HDAC8; incubate at 30°C for 60 min.
