About Jak/STAT Signaling

Mechanism of Jak/STAT Signaling
Jak/STAT signaling is initiated by the binding of cytokines or growth factors to their cognate receptors on the cell surface. Subsequent activation of these receptors triggers the recruitment and activation of Janus kinases (Jaks) associated with the cytoplasmic domain of the receptors. Activated Jaks then phosphorylate specific tyrosine residues on the receptors, creating docking sites for cytoplasmic STAT proteins. Phosphorylated STATs form dimers, translocate to the nucleus, and regulate the transcription of target genes, thereby modulating various cellular functions.
Moreover, the pathway is tightly regulated by negative regulators including suppressors of cytokine signaling (SOCS) and protein inhibitors of activated STATs (PIAS), which help maintain cellular homeostasis by inhibiting excessive or prolonged signaling.
