What is SPARCL1 protein?

SPARCL1 gene is a protein coding gene which situated on the long arm of chromosome 4 at locus 4q.22 that spans 57.7 Kb gDNA. SPARCL1 (Secreted Protein, Acidic and Rich in Cysteines­like 1), also known as hevin, SC1 or MAST9 is encoded by the SPARCL1 gene. It is a member of the SPARC family of extracellular glycoproteins that contained 664 amino acids (aa). Its molecular mass is approximately 75 kDa but migrates at ~130 kDa, which has been explained either by disulfide­linked homodimerization or by glycosylation and high acidity. The subcellular locations for SPARCL1 is usually secreted, or in extracellular space and extracellular matrix. SPARCL1 is an anti­adhesive protein that is highly expressed in lymph node, brain, heart, lung, skeletal muscle, ovary, small intestine, and colon, with lower levels in placenta, pancreas, testis, spleen, and thymus, and no expression in kidney, liver, and peripheral blood leukocytes. Human SPARCL1 contains a 16 aa signal sequence and a 648 aa mature region with four domains: a 416 aa N­terminal acidic region, a 23 aa follistatin­like domain, a 55 aa kazal­like segment and a 48 aa EF­hand/calcium­binding domain. Human mature SPARCL1 shares 67%, 69%, 78%, 76%, 72% and 72% aa identity with mouse, rat, equine, canine, porcine and bovine SPARCL1, respectively. The follistatin­like, kazal­like and calcium­binding domains of SPARCL1 show 61% aa identity with corresponding regions of SPARC.

What is the function of SPARCL1 protein?

SPARCL1 inhibits adhesion and spreading on a variety of substrate. It is thought to cause antiadhesive signaling that terminates neuronal migration, consistent with production by glial and neuronal cells during development or in response to trauma. SPARCL1 specifically promotes the formation and maintenance of thalamocortical excitatory synapses, by linking presynaptic neurexin-1α and postsynaptic neuroligin-1B. Importantly, SPARCL1 has been implicated in resilience to stress, showing an antidepressant-like effect in a model of social defeat. Also, SPARCL1 is one of the most downregulated transcripts in the frontopolar cortex of depressed/suicide victims. In tonsillar high endothelial venules (HEV), SPARCL1 may induce endothelial cell dissociation, promoting extravasation. SPARCL1 binds collagen in mice, deletion of SPARCL1 causes dermal collagen fibrils that are smaller in diameter and deficient in decorin.

SPARCL1 Related Diseases

Neurodegenerative Disorders. Downregulated SPARCL1 has been found in the cerebrospinal fluid (CSF) of individuals with Parkinson´s and Alzheimer´s diseases.

Emotional Control Disorder. SPARCL1 has been implicated in resilience to stress, also, SPARCL1 is one of the most downregulated transcripts in the frontopolar cortex of depressed/suicide victims. Tumors. SPARCL1 is a matricellular protein with anti-adhesive, anti-proliferative and anti-tumorigenic functions and is frequently downregulated in tumors such as colorectal carcinoma or non-small cell lung cancer.

Biomedical Application of SPARCL1 Protein

SPARCL1 has also been looked at as a potential biomarker of neurodegenerative disorders and has been found in the cerebrospinal fluid (CSF) of individuals with Parkinson´s and Alzheimer´s diseases. Also, SPARCL1 is a novel prognostic biomarker and correlates with tumor microenvironment in colorectal cancer or prostate cancer.One research reavealed SPARCL1 could act as a biomarker of maladaptive right ventricular remodelling in pulmonary hypertension.
