Auramine O fluorescence staining is a cold, non-heating method for acid-fast bacteria. After staining and counterstaining, specimens are examined under a fluorescence microscope equipped with a UV light source.
Acid-fast bacilli fluoresce bright yellow, while background material appears dark yellow. This method allows rapid screening at low magnification.
