Acid-fast bacteria contain mycolic acids in their cell walls, making them resistant to conventional staining. The modified Kinyoun method is a cold staining technique that does not require heating.
At room temperature, carbol fuchsin binds to mycolic acids, and after methylene blue counterstaining, acid-fast organisms remain red while the background appears blue. This method produces lighter staining than Ziehl–Neelsen but is suitable for weakly acid-fast organisms.
