Acid-fast bacteria contain lipid-rich cell walls that resist conventional staining. Ziehl–Neelsen staining is a WHO-recommended hot staining method.
Under heating conditions, carbol fuchsin penetrates the bacterial cell wall. After counterstaining with methylene blue, acid-fast bacteria remain red, while background and non-acid-fast bacteria appear blue. This method provides deeper staining and clearer contrast than the Kinyoun cold method.
