Significance: Acid protease (ACP) is a protein-hydrolyzing enzyme active under acidic conditions. It is widely used in alcohol fermentation, brewing, leather processing, fruit wine clarification, soy sauce production, and animal feed applications.
Principle: Under acidic conditions, ACP catalyzes the hydrolysis of casein, releasing tyrosine. Under alkaline conditions, tyrosine reduces phosphomolybdic acid complexes to produce tungsten blue, which exhibits a characteristic absorbance peak at 680 nm. ACP activity is calculated based on the increase in absorbance.
