Significance of Measurement: Alkaline Phosphatase (AKP/ALP) is a zinc-containing glycoprotein enzyme widely distributed in human tissues, with high abundance in the liver. It hydrolyzes phosphate monoesters under alkaline conditions.
Principle of Measurement: AKP/ALP catalyzes the hydrolysis of disodium phenyl phosphate to release phenol, which reacts with 4-aminoantipyrine and potassium ferricyanide to form a red quinone derivative. The increase in absorbance at 510 nm is used to calculate enzyme activity.
