Alkaline Phosphatase is widely distributed in mammalian tissues, particularly at sites of active transport. Under alkaline conditions (pH 9.2–9.8), ALP hydrolyzes a substrate to release naphthol, which couples with a diazonium salt to form an insoluble colored precipitate localized in the cytoplasm. This method is suitable for blood smears, bone marrow, cell smears, and frozen sections and provides accurate localization of enzymatic activity.
