Alkaline Phosphatase (ALP) is a phosphomonoesterase widely distributed in mammalian tissues, with optimal activity at pH 9.2–9.8. It is enriched at sites of active transport, including intestinal brush borders, renal proximal tubules, bile canaliculi, vascular endothelium, and smooth muscle cell membranes.
This ALP staining kit employs a modified Gomori calcium–cobalt method using sodium β-glycerophosphate as a substrate. Enzymatic hydrolysis releases phosphate ions, which are captured by metal ions to form an insoluble precipitate that is subsequently converted into a stable brown-black deposit. The method provides cascade signal amplification and allows qualitative or semi-quantitative detection of ALP activity in frozen and paraffin sections.
