Amyloid is an amorphous extracellular eosinophilic material that accumulates in various tissues and organs, resulting in amyloidosis. Amyloid is primarily composed of proteins arranged in antiparallel β-pleated sheet structures. Electron microscopy reveals non-branching fibrils arranged extracellularly in random orientations.
Traditional amyloid detection methods include methyl violet staining, Congo red staining, and polarized light microscopy. This kit employs a fluorescence-based method using Thioflavin T (ThT), a benzothiazole dye that exhibits enhanced fluorescence upon binding to amyloid fibrils and amyloid-β protein, enabling specific localization of amyloid deposits.
