Significance of Measurement: Pyruvate kinase (PK) is a key glycolytic enzyme that catalyzes the conversion of phosphoenolpyruvate to pyruvate, generating ATP. PK activity reflects glycolytic flux and cellular energy metabolism.
Principle of Measurement: PK catalyzes ATP generation, which is coupled with lactate dehydrogenase-mediated oxidation of NADH to NAD⁺. The decrease in absorbance at 340 nm corresponds to PK activity.
