Significance: Blood phosphorus mainly refers to inorganic phosphorus present as inorganic phosphate salts. Serum calcium and phosphorus concentrations are closely related, and when expressed in mg/dL, their product ([Ca] × [P]) normally ranges from 30 to 40. Values above 40 lead to calcium-phosphate deposition in bone, while values below 35 impair bone mineralization. Stable blood calcium and phosphorus levels are regulated by absorption, excretion, mineralization, and demineralization processes under hormonal control.
Principle: After removal of organic phosphorus from serum, inorganic phosphate reacts with ammonium molybdate to form phosphomolybdic acid, which is reduced by ferrous sulfate to form a blue complex with absorbance at 660 nm. Phosphorus content is calculated based on absorbance at 660 nm.
