The bacterial capsule is a polysaccharide layer external to the cell wall with low affinity for dyes. Traditional negative staining methods color the background while leaving the capsule unstained, but may result in misinterpretation.
This kit uses a classical capsule staining method with internal and external staining to enhance contrast and accuracy. Heat fixation is avoided to prevent capsule distortion. The method is suitable for identifying encapsulated bacteria such as Streptococcus pneumoniae, Haemophilus influenzae, Bacillus anthracis, and Clostridium perfringens.
