Alum hematoxylin alone is highly sensitive to acidic dyes, leading to nuclear fading during procedures such as Van Gieson or Masson trichrome staining.
This combined celestine blue and alum hematoxylin method enhances acid resistance. Ferric salts in celestine blue strengthen nuclear binding of hematoxylin, producing deeper nuclear staining that resists acid differentiation.
