Significance: CS (EC 2.3.3.1) is a key enzyme in the tricarboxylic acid cycle, widely present in mitochondria of animals, plants, microbes, and cultured cells, and serves as a rate-limiting step and major regulatory site.
Principle: CS catalyzes the condensation of acetyl-CoA and oxaloacetate to form citryl-CoA, which is further hydrolyzed to citrate. This reaction converts colorless DTNB to yellow TNB, which has a characteristic absorbance at 412 nm.
