Significance: NAD⁺ and NADH are essential coenzymes in glycolysis and the TCA cycle, mediating redox reactions that generate ATP and ROS. The NADH/NAD⁺ ratio reflects cellular respiratory activity and metabolic state. NAD⁺ degradation products also regulate signaling, metabolism, and gene expression.
Principle: NAD⁺ and NADH are extracted separately with acidic and basic solutions. NADH reduces oxidized MTT via PMS to form formazan, measured at 570 nm. NAD⁺ is first enzymatically reduced to NADH, followed by MTT reduction detection.
