Significance: NAD+ and NADH are essential coenzymes in glycolysis and the TCA cycle, mediating redox reactions that generate ATP and ROS. Principle: NAD+ and NADH are extracted separately with acidic and basic solutions. NADH reduces oxidized MTT via PMS to form formazan, measured at 570 nm. NAD+ is first enzymatically reduced to NADH, followed by MTT reduction detection.
