Amyloid is an extracellular eosinophilic substance lacking fixed morphology. Diseases characterized by amyloid deposition are collectively referred to as amyloidosis. Amyloid consists primarily of proteins arranged in antiparallel β-pleated sheet structures. Under electron microscopy, amyloid appears as non-branching fibrils arranged randomly in the extracellular space.
Histological methods for amyloid detection include methyl violet staining, Congo red staining, and polarized light microscopy. Traditional methyl violet staining shows low sensitivity and specificity. Congo red staining remains the classical and most reliable method. Following Highman’s modification, staining quality and contrast are significantly improved.
This kit contains Congo red solution and hematoxylin. Amyloid exhibits high affinity for Congo red through hydroxyl–amino group interactions, resulting in red staining.
