Amyloid is an extracellular eosinophilic material composed mainly of proteins arranged in antiparallel β-pleated sheet structures. Congo red staining is a classical and effective method for amyloid detection.
The modified Highman (methanol Congo red) method improves staining sensitivity and clarity. Amyloid exhibits strong affinity for Congo red due to hydrogen bonding between hydroxyl groups and amino groups, resulting in red staining. The kit typically includes Congo red solution and Mayer’s hematoxylin for nuclear counterstaining.
