Calcium-containing tissues are unsuitable for direct paraffin embedding due to density differences between calcium and paraffin. Such tissues should be fixed prior to or during decalcification.
Decalcification methods include organic acids, inorganic acids, and electrolytic techniques. The degree of decalcification should be monitored regularly to prevent over-decalcification, which may damage tissue structure and compromise staining quality.
