Elastic fibers are mainly distributed in arterial walls, alveolar walls, and skin, exhibiting strong refractivity and a yellow appearance when fresh. Common staining methods include Gomori aldehyde fuchsin, resorcin-fuchsin, orcein, Victoria blue, and iron hematoxylin.
Victoria blue is a triphenylmethane dye that selectively stains elastic fibers and hepatitis B surface antigen, with low background and good color stability.
The orcein-based elastic fiber staining method is simple and highly specific, although staining intensity may vary with reagent batches and may occasionally stain lipofuscin in hepatocytes.
