Significance of Measurement: Glutathione peroxidase (GPx, EC 1.11.1.9) is an important antioxidant enzyme that reduces hydrogen peroxide and lipid peroxides using reduced glutathione. GPx plays a critical role in preventing oxidative damage to membranes and macromolecules.
Principle of Measurement: GPx catalyzes the reduction of peroxides, producing oxidized glutathione (GSSG), which is subsequently reduced back to GSH by glutathione reductase with concomitant oxidation of NADPH. The decrease in absorbance at 340 nm reflects GPx activity.
