Significance: Glutamate pyruvate transaminase (GPT, EC 2.6.1.2) is widely distributed in animals, plants, microorganisms, and cultured cells and plays an important role in amino acid metabolism. In mammals, elevated serum GPT activity reflects hepatocellular damage and is recommended by the WHO as a sensitive indicator of liver injury.
Principle: GPT catalyzes the transamination of alanine and α-ketoglutarate to form pyruvate and glutamate. The reaction is terminated by 2,4-dinitrophenylhydrazine, which reacts with pyruvate to form a hydrazone. Under alkaline conditions, the hydrazone exhibits a red-brown color with absorbance at 505 nm, enabling activity calculation.
