Significance: Glutathione peroxidase (GPX, EC 1.11.1.9) is a key peroxide-degrading enzyme widely present in organisms. GPX enzymes are classified based on their active centers, with selenocysteine-containing GPX being predominant in most species. GPX reduces hydrogen peroxide and lipid peroxides, playing a critical role in antioxidant defense. This assay specifically measures selenocysteine-dependent GPX activity.
Principle: GPX catalyzes the oxidation of reduced glutathione (GSH) to oxidized glutathione (GSSG) using organic peroxides as substrates. GSH reacts with DTNB to produce a compound with characteristic absorbance at 412 nm. The decrease in absorbance reflects GPX activity.
