Gram staining is a standard bacteriological method for bacterial classification. Following crystal violet staining and iodine mordant treatment, an insoluble crystal violet–iodine complex forms within the cell wall. Gram-positive bacteria retain this complex due to their thick peptidoglycan layer and appear purple. In contrast, Gram-negative bacteria possess thinner cell walls and lipid-rich outer membranes; decolorization dissolves the lipid layer and releases the dye complex, allowing counterstaining with safranin to produce a red appearance.
