Granule-bound starch synthase (GBSS, EC 2.4.1.21) is bound within starch granules and catalyzes starch chain elongation, playing a primary role in amylose synthesis.
GBSS catalyzes glucose transfer from ADP-glucose to starch primers, generating ADP. Pyruvate kinase, hexokinase, and glucose-6-phosphate dehydrogenase subsequently catalyze NADP+ reduction to NADPH. NADPH formation at 340 nm is proportional to GBSS activity.
