The high iron diamine (HID)–Alcian blue method differentiates sulfated acidic mucins from sialomucins. Under strongly acidic conditions (pH < 2.5), sulfate groups ionize and bind to the cationic components of HID, producing brown-purple to brown-black staining.
This kit is used to distinguish sulfated acidic mucins and sialic acid–containing mucins in intestinal epithelium and metastatic tumors. It is often combined with AB–PAS staining for mucin subtype classification.
