Alcian blue is a copper phthalocyanine cationic dye that binds to acidic tissue components. It forms insoluble blue complexes with anionic groups such as carboxyl and sulfate groups present in acidic mucopolysaccharides. The dye consists of a copper-containing phthalocyanine core linked to four isothiouronium groups via thioether bonds, enabling strong electrostatic interactions with polyanions in tissues.
This hyaluronic acid staining method combines hyaluronidase digestion with Alcian blue staining. Hyaluronidase cleaves glycosidic bonds in hyaluronic acid and chondroitin sulfate. Loss of Alcian blue staining after enzymatic pretreatment indicates the presence of hyaluronic acid or chondroitin sulfate, whereas unchanged staining suggests their absence.
