Significance of Measurement: β-Galactosidase (β-GAL, EC 3.2.1.23) is widely present in animals, plants, microorganisms, and cultured cells. It catalyzes the hydrolysis of β-galactosidic bonds and also exhibits transgalactosylation activity. In plants, β-GAL plays an important role in energy release, polysaccharide metabolism, cell wall remodeling, and senescence-associated cell wall degradation.
Principle of Measurement: β-GAL hydrolyzes p-nitrophenyl-β-D-galactopyranoside to produce p-nitrophenol, which exhibits a maximum absorbance at 400 nm. The increase in absorbance over time is used to calculate β-GAL activity.
