Lysosomal β-galactosidase staining detects acid β-galactosidase activity within lysosomes and is commonly used as a reference control in cellular senescence studies. The enzymatic activity can be directly observed in cells or tissues under a standard light microscope.
This kit is suitable for both cultured cells and tissue sections. Most normal cells exhibit relatively high lysosomal β-galactosidase activity, making this staining useful as a reference for senescence-associated β-galactosidase assays. Senescence-associated β-galactosidase is expressed only during cellular aging, whereas lysosomal β-galactosidase is expressed under nearly all conditions.
Using X-Gal as the substrate, acid β-galactosidase catalyzes the formation of a dark blue product, allowing easy visualization of β-galactosidase-positive cells or tissues under a light microscope.
