Connective tissue contains collagen, reticular, and elastic fibers, among which collagen fibers are the most abundant and widely distributed. Masson’s trichrome staining is a classic and authoritative method for connective tissue visualization, particularly for collagen fibers. The technique relies on differences in molecular size and tissue permeability of anionic dyes, allowing selective staining of muscle fibers and collagen. After staining, muscle fibers appear red, while collagen fibers appear green or blue. This method is commonly used to distinguish collagen from muscle fibers and provides stable staining, short differentiation time, vivid color contrast, broad applicability to paraffin and frozen sections, and long-term color retention.
