The Albert staining method is commonly used for demonstrating metachromatic granules in Corynebacterium diphtheriae. The bacteria are Gram-positive rods that often arrange in V-, L-, or Y-shaped patterns.
After staining, metachromatic granules appear dark blue, while bacterial cytoplasm appears light blue, providing clear contrast.
