GMS staining is a classical method for basement membrane visualization. Oxidation exposes aldehyde groups in basement membrane mucopolysaccharides, which reduce methenamine silver to metallic silver. Gold chloride enhances contrast and stabilizes staining.
This method is extensively used in renal pathology to assess glomerular basement membrane alterations such as rupture, thickening, folding, and proliferation. After staining, basement membranes appear black against a green background.
