Significance: Acetylcholinesterase (AchE) is a serine hydrolase widely distributed in animal tissues and serum. It catalyzes the hydrolysis of acetylcholine and plays an essential role in regulating neurotransmission.
Principle: AchE hydrolyzes acetylcholine to generate choline, which reacts with 5,5′-dithiobis-(2-nitrobenzoic acid) (DTNB) to produce 5-thio-2-nitrobenzoic acid (TNB). TNB exhibits a characteristic absorbance at 412 nm, and the rate of absorbance increase is proportional to AchE activity.
