Significance of Measurement: Glutamate oxaloacetate transaminase (GOT, EC 2.6.1.1) is widely distributed in animals, plants, microorganisms, and cultured cells. It catalyzes reversible transamination reactions and plays an essential role in amino acid metabolism. GOT is highly abundant in cardiomyocytes and is commonly used as an auxiliary clinical indicator for myocardial infarction and myocarditis. Serum GOT levels also increase during liver injury.
Principle of Measurement: GOT catalyzes the transamination of α-ketoglutarate and aspartate to form glutamate and oxaloacetate. Oxaloacetate spontaneously decarboxylates to pyruvate, which reacts with 2,4-dinitrophenylhydrazine to form a colored hydrazone. Under alkaline conditions, the product exhibits a brown-red color. GOT activity is calculated by measuring absorbance at 505 nm.
