Significance: Nitric oxide (NO) is widely distributed in the nervous, cardiovascular, respiratory, digestive, and urogenital systems, with particularly high abundance in neural tissues. As a signaling molecule, NO plays important roles in physiological and pathological processes.
Principle: NO is readily oxidized to nitrite (NO2-) in vivo or in aqueous solutions. Under acidic conditions, nitrite reacts with sulfanilamide to form a diazonium compound, which subsequently couples with N-(1-naphthyl)ethylenediamine to produce a chromophore exhibiting a characteristic absorbance at 550 nm. Sample blanks are required to eliminate interference from endogenous pigments.
