Significance: Mitochondrial Complex I (EC 1.6.5.3), also known as NADH–CoQ reductase or NADH dehydrogenase, is the largest protein complex in the mitochondrial inner membrane. It catalyzes electron transfer from NADH to coenzyme Q and is a major site of superoxide generation in the respiratory electron transport chain. Its activity reflects both ETC function and reactive oxygen species production.
Principle: Complex I catalyzes the oxidation of NADH to NAD+. The rate of NADH oxidation is determined by monitoring the decrease in absorbance at 340 nm.
