Collagen fibers are the most abundant fibers in connective tissues and are widely distributed across organs. Type I collagen predominates in bone, skin, and tendons; type II in cartilage; type III in embryonic tissues, blood vessels, and gastrointestinal tract; and type IV in basement membranes. Aniline blue is a high-molecular-weight acidic dye that binds strongly to basic groups in collagen molecules.
This modified aniline blue staining kit consists of iron hematoxylin, PAB staining solution, and a weak acid solution, without picric acid. It is used for studying pathological alterations of collagen fibers. Under light microscopy, collagen fibers appear blue or blue-green, with yellow background staining.
