In this method, collagen fibers stain blue, muscle fibers and cytoplasm red, and nuclei blue-brown. Compared with conventional Masson staining, the modified protocol uses light nuclear staining to enhance contrast. It features short differentiation time, vivid coloration, broad compatibility with paraffin and frozen sections, and long-term color stability. The protocol avoids hazardous fixatives and improves operational safety.
