Cartilage consists of chondrocytes and extracellular matrix and is classified into hyaline, elastic, and fibrocartilage based on fiber composition. In the modified safranin O–fast green method, basophilic cartilage matrix binds safranin O and appears red, while acidophilic bone tissue binds fast green and appears green or blue. Safranin O staining intensity correlates with proteoglycan content and distribution. Cartilage degeneration leads to reduced or absent staining, which can be quantitatively analyzed using image analysis software. Proper differentiation is critical to avoid over- or under-staining.
