Monoamine oxidase (MAO), including MAO-A and MAO-B, is a flavoprotein bound to the outer membrane of mitochondria and catalyzes the oxidative deamination of neurotransmitters and biogenic amines. MAO activity is closely associated with organismal aging and is widely regarded as a biomarker of aging. It is predominantly distributed in various organs of vertebrates, particularly in secretory glands, brain, and liver. Significant changes in MAO activity have been detected in depression, oxidative stress-induced myocardial injury, and tissue fibrosis.
MAO activity is also present in invertebrates and plants such as legume sprouts, where it participates in monoamine metabolism at relatively low levels but still plays important physiological roles.
The Monoamine Oxidase (MAO) Staining Kit (NBT method) consists mainly of MAO fixative and MAO staining solution. Using tryptamine salts as the substrate and nitroblue tetrazolium (NBT) as the hydrogen acceptor, the hydrogen acceptor is reduced in situ at the enzyme active site during oxidative deamination, forming an insoluble blue formazan precipitate.
