Pathogenic nematodes can be isolated from diseased tissues by direct picking, funnel separation, or tissue homogenization. Live nematodes may be mounted for preliminary morphological observation, whereas detailed taxonomic analysis requires killing, fixation, optional staining, and preparation of permanent slides.
This conventional dehydration kit consists of two reagents primarily composed of glycerol and ethanol. Complete processing from fixation to mounting requires 12–18 hours. Fixed and dehydrated nematodes are suitable for long-term preservation.
