Neuronal cell bodies contain a large nucleus with sparse chromatin and a prominent nucleolus. The cytoplasm contains Nissl substance, which represents rough endoplasmic reticulum and ribosomes, appearing as basophilic granular structures. Nissl bodies can be demonstrated using basic dyes such as neutral red, methylene blue, toluidine blue, and methyl violet.
Nissl bodies are triangular or oval cytoplasmic aggregates found in neuronal soma and dendrites but absent from axons and axon hillocks. Their morphology, quantity, and distribution vary among neuron types and physiological states. Nissl substance is a key site of protein synthesis and decreases markedly following neuronal injury.
This Nissl staining kit features simple operation, stable staining, and short differentiation time, and is suitable for paraffin sections. Loss or dissolution of Nissl bodies is an important indicator of neuronal damage, such as encephalitis, ischemia, or axonal reaction.
